PROTEIN SCIENTIST - CHEMICAL BIOLOGY Cambridge, MA Job # RES002052
We are seeking an innovative and experienced protein scientist to join the Merck Exploratory Science Center (MESC) in Cambridge, MA.
The successful candidate will become part of a lab-based, creative and fast-paced team that will discover novel biology and invent impactful technologies targeting these areas:
Interaction between the microbiome and human cells/tissue, including the immune system
Host-pathogen interactions in important infectious diseases
Approaches that will impact other key areas of focus within Merck, such as oncology, inflammation, cardiovascular, metabolism, and neuroscience

Responsibilities include and are not limited to the following:
Manage the maintenance and daily operation of a Thermo LC and Q-Exactive HF Orbitrap
Develop and execute nanoflow LC-MS/MS workflows for discovery proteomics focused on identifying and characterizing novel protein-protein and protein-ligand interactions
Investigating potential therapeutic targets by designing and performing biochemical and cellular experiments that will be analyzed using LC-MS/MS
Characterizing protein bioconjugates to determine labeling ratios and identify sites of conjugation, such as Drug-to-Antibody Ratio (DAR) Calculations for Antibody-Drug Conjugates (ADCs)
Protein-small molecule binding-site elucidation
Developing novel techniques and solutions to challenging scientific questions
The candidate will be part of a cross-functional team contributing to the strategic direction of research programs across multiple therapeutic modalities
Act as a leader from the bench, driving experimentation design and execution
Provide scientific mentoring and technical training of team members as appropriate
Ensure all laboratory safety protocols are followed
Required: M.S. in Biochemistry, Protein Sciences, Chemical Biology, Biomedical Sciences or related discipline
Minimum of (5) years Academic or Pharmaceutical industry experience
Independent experimental design and execution of experiments
Rigorous scientific thinking and effective written and oral communication skills
Broad technical experience using LC/MSMS proteomics
Familiar with protein sample preparation, various protein labelling technologies, QC analysis, data processing, statistical analysis tools and protein identification
Experience using Thermo Orbitrap instrument(s)
Demonstrated critical thinking and trouble-shooting ability when handling challenging proteins such as aggregated protein, multicomponent complex, or membrane proteins
Targeted and quantitative bottom-up proteomics experience
Experience designing and performing biochemical and cellular experiments
Q Exactive LC-MS/MS experience
Experience with activity and affinity-based chemoproteomics
Experience with post-translation modifications (eg. Glycosylation, lipidation, etc)
